Bioavailability for humans of deuterium-labeled monoglutamyl and polyglutamyl folates is affected by selected foods

Dietary folate exists mainly as pteroylpolyglutamates (PteGlu sub n )5 which require deconjugation to pteroylmonoglutamates (PteGlu sub 1 ) by a Zn-dependent petroylpolyglutamate hydrolase (PPH), associated with the jejunal brush border membrane, prior to intestinal absorption. Although brush border PPH activity appears to exceed absorptive capacity as measured in vitro (Halsted 1990), significant reduction of PPH activity by dietary inhibitors, changes in intestinal pH, or disease may render PPH rate-limiting in the absorption of polyglutamyl folates. As recently reviewed (Gregory 1995), studies of the bioavailability of polyglutamyl folates in humans have indicated a bioavailability of ==75% (range ==50-100%) relative to the monoglutamyl form.
