At the 52nd ASMS conference, Bruker Daltonics, an operating company of Bruker BioSciences Corporation (NASDAQ: BRKR), today announces its new Proteineer-LC, a new LC-ESI-MALDI system for LC-based expression proteomics workflows. Moreover, Bruker Daltonics also introduced the new Top-Down Proteineer (TopPro(TM)) capability on its apex-Qe(TM) Q-q-FTMS system, allowing identification, primary structure determination and localization of post-translation modifications (PTMs) directly at the protein level, without the need for enzymatic digests.
Proteineer-LC
Proteineer-LC combines multi-LC-MS/MS proteomics approaches, e.g. using ESI ion trap mass spectrometry, with a new optimized sample splitting and MALDI fraction collection device, called Proteineer-FC. This enables a seamless LC-MALDI-MS/MS proteomics workflow with our TOF/TOF systems at high sensitivity, using our patented Anchorchip sample plate technology.
Information-dependant workflows can be utilized so that tentative protein IDs can be confirmed in an intelligent, results-driven way by our WARP-LC(TM) software. WARP-LC can analyze LC-MS(n) results from our HCTplus high capacity ion trap on the fly and then direct targeted MALDI-TOF/TOF experiments, based on the initial database search results. The combined information significantly increases ID success rates, allows identification of lower abundance proteins and enables further PTM characterization without LC time constraints using the LC-MALDI-TOF/TOF workflow.
Top-Down Proteineer
The unique Top-Down Proteineer (TopPro(TM)) is now available on the apex-Qe(TM) hybrid Q-q-FTMS system launched earlier this year. It combines the analytical power of FTMS equipped with a hybrid ESI-Q-q interface to perform "top-down" analysis of intact proteins in the FTMS analyzer cell, thus avoiding digestion and additional separation steps. The enormous resolving power of the FTMS is ideal for measuring protein fragment ions generated directly with electron-capture dissociation (ECD) in the FTMS analyzer cell, and can deliver high fidelity long sequence runs, as well as information on the localization of PTMs. Our unique ability for efficient selective ion enrichment using selected ion injection via the analytical quadrupole of the Q-q-FTMS extends the overall systems dynamic range and sensitivity by one to two orders of magnitude. MS Biotools software handles the database searching interface and confirms sequence annotations on the fly.
