As cytotoxics comprise one-third of the pipeline, they will remain the backbone of treatment. - the author believes Genentech/Roches Avastin (bevacizumab) will earn blockbuster status within the breast cancer market. Unlike Herceptin, Avastin is not limited to HER-2 positive tumors and will have a major impact on first-line metastatic disease. Avastin will dominate the breast cancer market with 2016 sales approaching $1.5bn. - GlaxoSmithKlines Tykerb (lapatinib) will expand treatment options for HER-2 positive, Herceptin-refractory, locally advanced and metastatic patients, with approval likely by early-2007. As an estimated 40% of patients are Herceptin-refractory, Tykerbs sales could reach over $500m by 2016 in this setting alone.
